What is the best way to give a user no-access to a directory?

I have a directory looks like this:

drwxr-xr-x 6 root root 1024 Sep 28 20:05 vol01/

how can i setup a new user (named "myuser") to have no access at all to
/vol01 , not even read access.?

I am trying to not change the perms on /vol01 if i dont have to since alot
of other users/groups need access.
