Here are a few that come quickly to mind. And I apologize if these are basic tips, but sometimes the simple things get overlooked.
* Be sure you research if there are any required APAR's that will be needed on any application/compiler you now run to successfully run in the 5.x environ. For example: (1) C for AIX 5 requires IY34533, IY34534, IY34535, and IY34623; or (2)Oracle 8.i required a patch 2896876 is required to run it in 64-bit mode. Obviously, you will need to do this leg work for your individual applications.
* I've also heard about problems migrating with a mirrored rootvg, and that you should unmirror it first.
* Update all you firmware and microcodes prior to 5.x
* they've changed the utmp and wtmp structure so that (for example in my case) the who command doesn't work when I log into the box using ssh. The work around is to recompile ssh. but at first I was a little concerned when I wasn't getting anything back from my 'who' command.
