When I try this (I'm trying to change my interface from 100Mb-Full
Duplex to Auto-negotiate), it tells me that characteristic can't be
changed. Do I need to make the device unavailable before making this

"Apply change to DATABASE only" make it equal to yes.

Which really only invokes the -P command the ultimate chdev command....
which:

-P Changes the device's characteristics permanently in the Customized
Devices object class without actually changing the device. This is
useful for devices that cannot be made unavailable and cannot be changed
while in the available state. The change can be made to the database
with the -P flag. By restarting the system, the changes will be applied
to the device. This flag cannot be used with the -T flag.
